For potable water samples, confirm with Lauryl Tryptose and Brilliant Green Bile Broth tubes. Samples that test positive for total coliform bacteria should be further analyzed for the presence of fecal coliform or E. coli using EC Medium or EC Medium with MUG respectively.

Dissolve 23g in one liter distilled water. Heat solution to boiling with continuous stirring for one minute. Sterilize by autoclaving at 121°C for 15 minutes. Mix medium with gentle agitation to avoid bubbles prior to pouring plates. Store at room temperature.

S2Media™ Yersinia Selective Agar (Schiemann's CIN Agar) is a selective and differential medium used for isolation and enumeration of Yersinia species from food samples. Its formulation incorporates multiple selective agents, sodium deoxycholate, crystal violet, cefsulodin, irgasan, and novobiocin, to inhibit enteric flora. Yersinia spp. characteristically ferment mannitol, producing a concentrated acid reaction in the colony center that creates a distinctive red “bull’s-eye” morphology.
